名称Mavoglurant
描述Mavoglurant (AFQ056) is a potent, selective, non-competitive, and orally active metabotropic glutamate receptor 5 (mGluR5) antagonist with an IC50 of 30 nM, demonstrating greater than 300-fold selectivity over 238 tested targets, and it is widely applied in research on Fragile X syndrome and L-dopa-induced dyskinesias in Parkinson’s disease, while Mavoglurant incorporated alkyne functional group, which enables copper-catalyzed azide–alkyne cycloaddition click chemistry, facilitating target engagement and molecular tracking studies.
体外活性在表达人 mGluR5a 的 L(tk-) 细胞功能测定中，Mavoglurant 拮抗激动剂诱导的信号传导，抑制磷脂酰肌醇周转 (IC₅₀ = 30 nM) 和细胞内 Ca2+ 动员 (IC₅₀ = 110 nM)。放射性配体结合实验证实其与跨膜变构位点的相互作用，以 47 nM 的 IC₅₀ 置换 3H-AAE327 [1]。
体内活性在小鼠应激诱导的高热 (SIH) 模型中，口服给予 Mavoglurant (0.1 - 10 mg/kg) 以剂量依赖的方式减轻了应激反应。在大鼠药代动力学分析中，该化合物显示出脑渗透性；静脉注射 (3.1 mg/kg) 后，脑部 Cmax (8400 pmol/g) 高于血浆 (3330 pmol/mL)。在大鼠口服给药 (9.4 mg/kg) 实验中，生物利用度为 32%，末端半衰期 (T1/2) 为 2.9 小时 [1]。
