描述CGP-42112 acetate is a potent angiotensin receptor AT2 agonist that inhibits the increase in protein kinase A activity produced by LPS.
体外活性在浓度≥1 nM时，CGP-42112 acetate显著地抑制了从基础值开始的cGMP产生，并且还抑制了从基础值开始的TH-酶活性。CGP-42112 acetate对于TH-酶活性和cGMP产生的抑制作用被PD123319（AT(2)-R拮抗剂）所抵消，而CV-11974（AT(1)-R拮抗剂）效果不显著[1]。[125I]CGP-42112 acetate选择性地结合到AT2血管紧张素II受体亚型。其结合亲和力在大脑中高于在肾上腺中。Beta-巯基乙醇可增强[125I]CGP-42112 acetate在大脑中的结合，但不改变其在肾上腺中的结合[2]。[125I]CGP-42112 acetate（Kd = 0.07-0.3 nM，依据研究区域而异）的高亲和力结合特异性地针对AT2受体。这种选择性通过AT1配体洛沙坦无法竞争以及AT2配体PD 123177和未标记的CGP-42112 acetate以及非选择性肽Ang II和血管紧张素III（Ang III）的竞争来展示[3]。
体内活性CGP-42112 acetate（0.1及1 mg kg^-1 min^-1）和PD 123319（0.36及1 mg kg^-1 min^-1）的静脉输注将脑血流(CBF)自动调节的上限向更高的血压水平移动，而不影响基线CBF[4]。
