名称UP158
描述UP158 is a small molecule activator of valosin-containing protein (VCP), activating VCP function by enhancing ATPase activity with a half-maximal effective concentration (EC₅₀) of 2.57 μM. VCP (also known as p97) is a critical AAA+ ATPase involved in protein degradation and cellular homeostasis regulation. Therefore, UP158 serves as a VCP activation tool compound primarily used to study the functions of VCP in protein quality control, endoplasmic reticulum-associated degradation (ERAD), and neurodegenerative diseases such as multisystem proteinopathy.
体外活性方法：将纯化重组野生型VCP与不同浓度UP158孵育并检测ATPase活性，同时以25 μM UP158进行ATP动力学实验。 结果：UP158激活VCP ATPase活性，EC50为2.57 μM，最大激活幅度约50%；25 μM UP158使VCP的kcat提高超过50%。[1]
