Bioactivity

名称MPTP
描述MPTP is a dopaminergic neurotoxic substance capable of penetrating the blood-brain barrier, commonly used to establish Parkinson's disease animal models. As an upstream precursor of MPP⁺, MPTP further mediates neuronal apoptosis.
体外活性方法:以小鼠膈神经 - 膈肌标本为研究模型，采用 50 mM 4 - 苯基吡啶进行预处理，分别测定 MPTP 与右旋筒箭毒碱抑制抽搐振幅的半数抑制浓度 IC₅₀，对比预处理前后的药效差异。 结果:经 50 mM 4 - 苯基吡啶预处理后，小鼠膈神经 - 膈肌标本中，MPTP 抑制抽搐振幅的 IC₅₀由 53 mM 降至 18 mM；d - 筒箭毒碱的 IC₅₀由 0.7 mM 降至 0.3 mM，提示 4 - 苯基吡啶可明显增强两种药物的抑制活性 [2]。
体内活性方法:选用 MPTP 建立帕金森病动物模型，依托该化合物诱导的神经损伤特征，探究多巴胺能神经元变性、线粒体功能异常及神经炎症的病理机制；同时检测 MPTP 体内代谢过程，分析其代谢产物 MPP + 在羊血清中的半衰期。 结果:MPTP 是构建帕金森综合征动物模型的经典造模试剂，可模拟自然病理状态下的神经退行性病变，适用于多巴胺能神经元损伤、线粒体功能障碍与神经炎症相关机制研究。MPTP 进入机体后可快速代谢转化为 MPP⁺，其中 MPP⁺在羊血清中的半衰期约为 6 天。
