Bioactivity

名称ACT-451840
描述ACT-451840 is capable of eliminating both sensitive and resistant strains of Plasmodium falciparum and has the advantages of oral activity, potency, low toxicity, rapid onset of action in all asexual blood stages of Plasmodium, and a mechanism of action similar to that of artemisinin derivatives. In in vivo experiments, ACT-451840 showed excellent antimalarial effects at a dose of 30 mg/kg, with a parasite reduction rate of 99.80%.
体外活性ACT-451840 对药物敏感的恶性疟原虫 NF54 菌株的 50% 抑制浓度为 0.4 nM；ACT-451840 可有效阻止雄配子从配子体阶段形成，50% 抑制浓度为 5.89 nM，并以剂量​​依赖性方式阻断蚊子卵囊的发育，50% 抑制浓度为 30 nM。[2]
体内活性体内药效模型中，ACT-451840对恶性疟原虫的 90% 有效剂量为 3.7 mg/kg，对伯氏疟原虫的 90% 有效剂量为 13 mg/kg。
