Bioactivity

名称BNS808
描述BNS808 is an orally active, selective CB1R antagonist with an IC50 of 0.8 nM, demonstrating significant selectivity for CB2R and minimal brain penetration. It is being studied for the treatment of obesity and related metabolic complications, such as metabolic dysfunction-associated steatotic liver disease (MASLD). BNS808 reduces drug exposure to the central nervous system, enhancing safety, and minimizes drug interactions through high plasma protein binding.
体外活性BNS808在HepG2细胞中显示出极低的细胞毒性，IC 50 为16.84 μM。BNS808的hERG IC 50 为5.39 μM，这表明对CB 1 R的IC 50 为0.8 nM，具有较低心脏毒性潜力。BNS808 (0.2 mg/mL, 10 分钟) 对CYP3A4表现出中度抑制，对CYP2C9和CYP2C19表现出弱中度抑制，对CYP1A2和CYP2D6则表现出弱抑制，其IC 50值分别为2.99、8.33、18.0和>50 μM。
体内活性在C57Bl/6小鼠中初步研究表明，单次口服给药BNS808 (1 mg/kg) 后1至8小时内，脑渗透性有限 (平均13.7 ng/g)。而在长期口服给药 (24天) 后，脑渗透性约为20 ng/g。在野生雄性C57Bl/6J小鼠中，无论是低剂量 (1 mg/kg) 还是高剂量 (10 mg/kg) 口服BNS808，均未观察到中枢神经系统介导的副作用，运动活动也无显著变化。同时，BNS808以1 mg/kg/天剂量口服24天可有效降低饮食诱导肥胖 (DIO) C57Bl/6J小鼠体重，并改善其代谢状态。
