Bioactivity

名称NF449
描述NF449 is a potent antagonist of the P2X1 receptor, displaying IC50 values of 0.28 nM, 0.69 nM, and 120 nM for rP2X1, rP2X1+5, and P2X2+3, respectively. It acts selectively against Gsα G proteins (G Protein). NF449 inhibits the binding rate of GTP[γS] to Gsα-s, suppresses the stimulation of adenylate cyclase activity, and blocks the coupling of β-adrenergic receptors with Gs.
体外活性NF449 抑制 GTP[γS] 和 rGsα-s 的结合速率，同时几乎不影响与 rGiα-1 的结合 (IC 50 =140 nM)，且通过外源添加的 Gsα-s 抑制 S49 环化膜（缺乏内源性 Gsα）中腺苷酸环化酶活性的提升，并阻断 β-肾上腺素能受体与 Gs 的偶联 (EC 50 =7.9 μM)。
体内活性在剂量为 10 mg/kg 时，NF449 能抑制由 5 g/mL 胶原蛋白在 WT 小鼠血小板中引发的离体聚集，而对 5 μM ADP 诱导的聚集无影响。在较高剂量 (50 mg/kg) 下，NF449 不仅能抑制血小板对 10 g/mL 胶原蛋白的反应，还能抑制对 5 μM ADP 的反应，这表明其非选择性地抑制 P2Y1 和/或 P2Y12 受体活性。
