Bioactivity

名称5α-Androstan-3α-ol
描述5α-Androstan-3α-ol is a γ-aminobutyric acid (GABA) receptor agonist. It inhibits the transcriptional activity of the orphan nuclear receptor CAR (constitutive androstane receptor). By binding to CAR and promoting its dissociation from coactivator proteins, 5α-Androstan-3α-ol negatively regulates CAR's activity. This compound is useful for studying hormone metabolism and drug detoxification mechanisms mediated by nuclear receptors.
体外活性方法： 大鼠海马脑片CA3区灌流55 μM 4-氨基吡啶（4-AP）诱导癫痫样放电，共灌流5α-Androstan-3α-ol（10、50、100 μM），使用细胞外场电位记录方法检测癫痫样放电频率变化。 结果： 5α-Androstan-3α-ol 浓度依赖性地抑制海马脑片CA3区癫痫样放电频率，100 μM时几乎完全消除放电[1]。
体内活性方法： 雄性NIH瑞士小鼠（25–30 g）腹腔注射5α-Androstan-3α-ol（10–300 mg/kg），在6 Hz电刺激（32 mA, 3 s）模型中，于给药后15 min进行电刺激，以10 s内恢复探索行为为保护终点，评价抗惊厥活性。 结果： 5α-Androstan-3α-ol 剂量依赖性地保护小鼠免受6 Hz电刺激诱导的惊厥，ED₅₀ = 29.1 mg/kg [1]。 方法： 雄性Sprague-Dawley大鼠侧脑室植入套管，通过远程控制输液泵于5 min内脑室内输注5α-Androstan-3α-ol硫酸盐（10 μg/10 μL，溶于10%乙醇生理盐水），输注期间及输注后30 min持续观察行为学反应，并同步记录皮层脑电图（EEG）[1]。 结果： 5α-Androstan-3α-ol硫酸盐诱导大鼠出现镇痛、湿狗样抖动、身体抽搐、僵直、Straub尾、活动过度、过度理毛、刺激超敏、攻击性、逃避行为、EEG棘波以及行为和EEG癫痫发作；部分反应持续数小时[1]。 方法： 雄性Sprague-Dawley大鼠侧脑室内输注5α-Androstan-3α-ol硫酸盐（10 μg/10 μL）前15 min，腹腔注射纳洛酮（5 mg/kg）或等量生理盐水，比较两组行为学及EEG反应差异。 结果： 纳洛酮预处理显著减少5α-Androstan-3α-ol硫酸盐诱导的EEG棘波、EEG癫痫发作、行为性癫痫、冰冻凝视、湿狗样抖动、站立行为和身体抽搐[2]。 方法： 雄性Sprague-Dawley大鼠侧脑室内输注5α-Androstan-3α-ol硫酸盐（10 μg/10 μL）或溶剂对照，输注后15 min采用热板法（44.5°C）测定后爪舔舐次数，评估镇痛效应。 结果： 与溶剂对照组相比，5α-Androstan-3α-ol硫酸盐处理组大鼠在15 min内后爪舔舐次数显著减少，表明具有抗伤害感受作用[2]。
