Bioactivity

名称Scoulerine HCl
描述Scoulerine HCl is a multi-target inhibitor with antibacterial, antitumor, and antioxidant activities. Scoulerine HCl targets GABA receptors and BACE1 and inhibits mitochondrial dehydrogenase activity, thereby suppressing cancer cell proliferation, migration, invasion, epithelial-mesenchymal transition, and stemness maintenance. Scoulerine HCl also disrupts microtubule structure by acting on the PI3K/Akt/mTOR signaling axis and α1D-adrenergic receptors, thereby inducing cell cycle arrest and apoptosis. Additionally, Scoulerine HCl regulates endoplasmic reticulum stress and mitochondrial function, and possesses various pharmacological activities, including anti-Plasmodium falciparum and antiemetic and antitussive effects.
体外活性方法：A549(肺癌)、A2780(卵巢癌)、SK-BR-3和MCF-7(乳腺癌)加入Scoulerine HCl(1, 5, 10, 20, 50 μM)，连续处理72小时，实时电阻抗监测细胞增殖。 结果：Scoulerine HCl呈现浓度依赖性抑制贴壁癌细胞增殖，10 μM 以上显著抑制。[1] 方法：Jurkat, MOLT-4白血病细胞加入加入Scoulerine HCl(2.5, 5, 10, 15, 20 μM)，连续处理24、48小时，台盼蓝拒染法计数活细胞。 结果：Scoulerine HCl呈现浓度和时间依赖性降低细胞活力和增殖。[1] 方法：OVCAR3细胞加入Scoulerine HCl (5, 10 μmol/L)，预处理48小时，处理后继续培养10天，结晶紫染色、分光光度法检测细胞克隆能力。 结果：Scoulerine HCl呈剂量依赖性降低细胞克隆形成能力，10 μmol/L时抑制率约78.3%。[2]
体内活性方法：为研究Scoulerine HCl的体内抗肿瘤疗效，选用BALB/c裸鼠，SK-OV-3细胞皮下移植瘤，肿瘤体积达0.52 mm³时开始给药Scoulerine HCl(1, 2.5, 5, 10 mg/kg)，至实验结束。 结果：Scoulerine HCl呈剂量依赖性减小肿瘤质量和体积，10 mg/kg时肿瘤质量从约2.1 g降至约0.7 g，体积从约1800 mm³降至约600 mm³。[2]
