名称Zinpyr-1
描述Zinpyr-1 is a zinc ion-responsive fluorescent indicator and cell membrane-permeable metal-binding probe. It binds to Mn²⁺ to form a complex with enhanced fluorescence signals. Moreover, Zinpyr-1 specifically binds to free zinc ions in serum, enabling quantitative determination of free zinc ion concentration. In plant systems, the fluorescence signal of Zinpyr-1 reflects the relative zinc ion level in root cells and is mainly localized in specific regions of root cells. It can be applied to the screening of plant zinc transporter mutants and research on zinc homeostasis regulation.
体内活性方法：采用 Zinpyr-1 与 Mn²⁺构建复合物，在不同 pH 条件下检测其对 Zn²⁺的荧光响应特性；通过细胞实验评价其膜通透性与细胞毒性；并在血清体系及拟南芥幼苗模型中验证锌离子检测与成像能力。 结果： 1.Zinpyr-1(10 μM，pH 6–8)可与 Mn²⁺形成 1:2 复合物，用于 Zn²⁺检测时荧光动态范围较游离配体提升 25 倍(pH 7.0 时为 106)；在 pH 6.0 和 8.0 下仍保持较高动态范围，且对 Zn²⁺的选择性显著优于体内常见一价、二价阳离子 。 2.Zinpyr-1(10 μM)与 Mn²⁺形成的复合物可穿透细胞膜，对 HeLa 细胞无毒性，背景荧光极低且结合 Zn²⁺后荧光显著激活，适用于活细胞锌离子成像。 3.Zinpyr-1(0.1 mM)与 Mn²⁺形成的复合物可作为锌响应型 MRI 造影剂使用 [1]。 4.Zinpyr-1 在血清中可稳定产生用于游离锌定量的最小荧光(Fmin)与最大荧光(Fmax)，最大激发波长 507 nm、最大发射波长 526 nm，可实现血清游离锌的可靠定量 [2]。 5.Zinpyr-1(5–20 μM，15 min–3 h)可作为拟南芥幼苗根细胞内相对锌水平的荧光探针，荧光强度与胞内锌浓度呈正相关 [3]。
