Litifilimab (also known as BIIB-059) is an investigational, humanized monoclonal antibody that targets blood dendritic cell antigen 2 (BDCA2), a receptor exclusively expressed on plasmacytoid dendritic cells (pDCs). By binding to BDCA2, litifilimab triggers the internalization and downregulation of the receptor, leading to the inhibition of pDC activation and the subsequent reduction of type I interferon (IFN) production. Since pDCs are a major source of pathogenic type I IFN in systemic lupus erythematosus (SLE) and other autoimmune diseases, this mechanism aims to dampen a key driver of autoimmunity. It is being clinically evaluated for the treatment of cutaneous lupus erythematosus (CLE) and systemic lupus erythematosus (SLE).
