Lenzilumab is a fully human monoclonal antibody that selectively targets and neutralizes granulocyte-macrophage colony-stimulating factor (GM-CSF), a pro-inflammatory cytokine. By binding to GM-CSF, it prevents the cytokine from interacting with its receptor (GM-CSFR) on immune cells, thereby inhibiting the downstream signaling cascade that drives hyperinflammation and cytokine release syndrome (CRS). It has been investigated as a therapeutic agent for conditions characterized by excessive immune activation, most notably in hospitalized patients with COVID-19 pneumonia to prevent or mitigate severe respiratory complications. It is also being explored in other hyperinflammatory and autoimmune contexts, such as acute graft-versus-host disease (aGVHD).
