Garivulimab (also known as BGB-A333) is an investigational, humanized immunoglobulin G1 (IgG1) monoclonal antibody being developed for cancer immunotherapy. It functions as a programmed death-ligand 1 (PD-L1) checkpoint inhibitor. By binding with high affinity to PD-L1 expressed on tumor cells and tumor-infiltrating immune cells, garivulimab blocks its interaction with the programmed cell death-1 (PD-1) receptor on T cells. This blockade releases the inhibitory "brake" that the PD-1/PD-L1 pathway places on T-cell activation and function, thereby restoring and enhancing the T-cell-mediated anti-tumor immune response. As an IgG1 antibody, it may also engage in potential Fc-mediated effector functions, such as antibody-dependent cellular cytotoxicity (ADCC) against PD-L1-expressing cells.
