Penpulimab (also known as AK‑105) is an investigational, fully human monoclonal antibody that targets programmed cell death‑1 (PD‑1), an inhibitory immune checkpoint receptor. By binding to PD‑1 on T cells, it blocks the interaction between PD‑1 and its ligands PD‑L1 and PD‑L2, thereby reversing PD‑1‑mediated immunosuppression and restoring anti‑tumor T‑cell activity. It is engineered with an IgG1 backbone that is Fc‑silent (modified to minimize antibody‑dependent cellular cytotoxicity and complement‑dependent cytotoxicity), focusing its mechanism on pure checkpoint blockade.
