Patritumab (also known as U3‑1287 or AMG‑888) is a fully human monoclonal antibody that targets human epidermal growth factor receptor 3 (HER3, ErbB3), a member of the EGFR/ErbB family. By binding to HER3, it blocks the interaction between HER3 and its ligand neuregulin (NRG), thereby inhibiting downstream signaling pathways (e.g., PI3K/AKT) that promote tumor cell survival and resistance to therapy. Unlike other ErbB family members, HER3 has impaired kinase activity and relies on heterodimerization (typically with HER2) for signaling.
