Omburtamab (also known as 8H9 or B7‑H3) is a murine monoclonal antibody that targets B7‑H3 (CD276), an immune checkpoint molecule belonging to the B7 family. B7‑H3 is overexpressed on a wide range of solid tumors, including neuroblastoma, brain tumors (e.g., diffuse intrinsic pontine glioma, medulloblastoma), and sarcomas, while its expression in normal tissues is limited. Omburtamab is not used as a naked antibody; it is radiolabeled with iodine‑131 (¹³¹I) for targeted radiotherapy. When administered directly into the tumor cavity or cerebrospinal fluid (intrathecal/intracavitary delivery), it delivers localized radiation to B7‑H3‑expressing tumor cells. It has been investigated for the treatment of leptomeningeal metastases and central nervous system tumors in pediatric and adult patients.
