Bemarituzumab (formerly FPA144) is a humanized monoclonal antibody of the IgG1 kappa isotype. It is an anticancer therapeutic designed with a dual mechanism of action. First, it functions as a signaling antagonist by binding to its target, thereby inhibiting the downstream tumor proliferation and survival pathways, such as MAPK and PI3K/AKT. Second, it is Fc-engineered (defucosylated) to enhance its affinity for the Fc gamma RIIIa (CD16a) receptor on Natural Killer (NK) cells, thereby boosting its Antibody-Dependent Cellular Cytotoxicity (ADCC) to directly destroy tumor cells. Bemarituzumab is primarily being investigated for the treatment of FGFR2b positive, unresectable locally advanced or metastatic gastric or gastroesophageal junction (G/GEJ) adenocarcinoma.
