Mipasetamab is a human IgG1-kappa monoclonal antibody that specifically targets the AXL receptor tyrosine kinase, a member of the TAM (TYRO3, AXL, MERTK) receptor family. As an IgG1 isotype, it is designed to engage immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC). AXL is often overexpressed or constitutively activated in a wide range of cancers. By binding to AXL, mipasetamab blocks its interaction with its ligand Gas6, inhibits downstream oncogenic signaling pathways (e.g., PI3K/AKT, MAPK), and may promote immune-mediated elimination of tumor cells. It is being developed as a therapeutic agent for AXL-dependent cancers, including non-small cell lung cancer (NSCLC), breast cancer, and pancreatic cancer, particularly in settings of therapeutic resistance.
