One unit is defined as the enzyme quantity which oxidizes one μ mole of L-a-Glycerophosphate per minute under the condition described below.
Reagents
A. Aminoantipyrine Solution (5 mg/mi)
Weigh 0.25 g of s-aminoantipyrinc and dissoive in 20 ml ol dcionized water. Fill up to 50ml with deionized water.
B. Phenol Solution (50 mg/ ml)
Weigh 5.0 g of phenol and dissolve in 30 ml of deionized water. Fill up to 100 ml with deionized water.
C. 0.2M Phosphate Buffer (KH2PO4-NaOH, pH 20)
D. 002M Phosphate Buffer (KH2PO4-NaOH, pH 20)
E. Aminoantipyrine-Phenol Solution
Dilute 6 ml of Aminoantipyrine Solution (A), 0.5 ml of Phenol Solution (B) and 200 purpurogallin units of Per-oxidase (produced by Amano Enzyme Inc.) with 0.2M Phosphate Buffer (C). Fill up to 50 ml with 0.2M Phosphate Buffer (C). (Expires after 14 days at 2-8°C)
F. Substrate Solution
Weigh 36.7 g of DL-α-glycerophosphate·2Na·5H2O (produced by Wako Pure Chemical Industries, Ltd. *) and dissolve in approx. 40 ml of deionized water. Adjust the pH to 7.0 with 4N HCl and fill up to 100 ml with deionized water. Then, filtrate the solution with 0.45 um membrane filter.
G. Diluent
Weigh 1.00 g of bovine serum albumin (produced by Wako Pure Chemical Industries, Ltd.: Fraction V*), 66.0 g of (NH,),SO, and dissolve in 002M Phosphate Buffer (D). Fil up to 1,000 ml with 0.02M Phosphate Buffer (D). (Expires after 1 month at 2-8°C)
H. Enzyme Solution
Weigh 20 mg of L-a-Glycerophosphate Oxidase "Amano" and dissolve in chilled Diluent (G).
Enzyme Solution should be prepared so that the value of △OD/ minute becomes in the range of 0.020±0.005
