研究背景：c-Kit 是一种酪氨酸蛋白激酶 (tyrosine-protein kinase)，作为细胞因子 KITLG/SCF 的细胞表面受体，在细胞存活与增殖、造血作用 (hematopoiesis)、干细胞维持、配子发生 (gametogenesis)、肥大细胞 (mast cell) 发育与功能以及黑色素生成 (melanogenesis) 中发挥关键作用。结合 KITLG/SCF 后，c-Kit 可激活多条信号通路，包括磷酸化 PIK3R1、PLCG1、SH2B2/APS 和 CBL，并通过磷酸化磷脂酰肌醇 3-激酶 (phosphatidylinositol 3-kinase) 的调节亚基 PIK3R1 激活 AKT1 信号通路。此外，c-Kit 还通过 GRB2 传递信号，激活 RAS、RAF1 及 MAP 激酶 MAPK1/ERK2 和/或 MAPK3/ERK1，并促进 STAT 家族成员 STAT1、STAT3、STAT5A 和 STAT5B 的激活。PLCG1 的激活可产生二酰基甘油 (diacylglycerol) 和 1,4,5-三磷酸肌醇 (inositol 1,4,5-trisphosphate) 等信号分子。c-Kit 信号受蛋白磷酸酶 (protein phosphatases) 及受体快速内化和降解的调控，其激活还能促进蛋白磷酸酶 PTPN6/SHP-1、PTPRU 以及转录因子 STAT1/3/5A/5B 的磷酸化。
