Biological Activity:

Inhibitor of Hedgehog signaling (IC50 = 500 - 700 nM); binds directly to Smoothened (Smo). Inhibits the response of target tissues to Sonic hedgehog signaling, possibly via the sterol sensing domain of Patched. Teratogenic.

Applications:   Has antibacterial properties. Jervine demonstrates teratogenic properties. It is the starting material for C-nor-D-homosteroids. Jervine induces holoprosencephaly and blocks endogenous Sonic hedgehog signaling.

Jervine is a steroidal alkaloid with molecular formula C27H39NO3 which is derived from the Veratrum plant genus. Similar to cyclopamine, which also occurs in the Veratrumgenus, it is a teratogen implicated in birth defects when consumed by animals during a certain period of their gestation.An alkaloid isolated from the corn lily (Veratrum californicum) is a Sonic Hedgehog gene pathway antagonist with potential anticancer properties. Jervine is a naturally occuring steroidal alkaloid that causes cyclopia by blocking sonic hedgehog signaling. This is derived from the Veratrum plant species. It is a close structural analog of cyclopamine which specifically inhibits the hedgehog (Hh) pathway by interaction with the hedgehog signaling protein Smo. Jervine can be used to induce abnormal morphogenesis in a number of experimental models. Jervine is an inhibitor of Smo.
