Cytokeratin 7 (CK7) is a member of the cytokeratin family, a group of intermediate filament proteins critical for maintaining epithelial cell structure and integrity. Expressed in various simple, glandular, and transitional epithelia, CK7 is commonly found in tissues such as the lung, breast, thyroid, and urogenital tract. Its antibody is widely utilized in diagnostic pathology to identify epithelial cell origins, particularly in cancer classification.

As a type II acidic keratin (paired with CK19), CK7 helps distinguish between adenocarcinomas of different primary sites. For example, CK7+/CK20− profiles often indicate lung or ovarian cancers, while CK7−/CK20+ patterns suggest colorectal origins. It also aids in differentiating carcinomas from mesotheliomas or sarcomas.

In research, CK7 antibodies are essential for studying epithelial differentiation, metastasis, and tissue regeneration. Immunohistochemistry (IHC) remains the primary method for detecting CK7 expression, though challenges like cross-reactivity or variable staining intensity require careful validation. Despite limitations, CK7 remains a cornerstone biomarker in tumor diagnostics, reflecting its reliability in tracing epithelial lineage and guiding clinical decision-making.
