ERK1 (Extracellular Signal-Regulated Kinase 1), also known as MAPK3. is a serine/threonine kinase within the mitogen-activated protein kinase (MAPK) family. It plays a central role in the RAS-RAF-MEK-ERK signaling cascade, which regulates critical cellular processes such as proliferation, differentiation, survival, and apoptosis. ERK1 is activated through dual phosphorylation at Thr202 and Tyr204 residues by upstream MAPK/ERK kinases (MEK1/2) in response to extracellular stimuli like growth factors, cytokines, or stress signals. Once activated, ERK1 translocates to the nucleus to phosphorylate transcription factors (e.g., c-Fos, c-Jun) or interacts with cytoplasmic targets to modulate cellular responses.

ERK1 antibodies are essential tools for studying this pathway in biomedical research. They are widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry to detect ERK1 expression, phosphorylation status (using phospho-specific antibodies), and subcellular localization. Commercial ERK1 antibodies are often developed against conserved epitopes, such as the C-terminal region, to ensure specificity across species (human, mouse, rat). Some antibodies distinguish between ERK1 (44 kDa) and its closely related isoform ERK2 (42 kDa). Validation includes knockout controls or siRNA-mediated silencing to confirm target specificity.
