The Phospho-Rb (Thr356) antibody is a specialized tool used to detect the retinoblastoma protein (Rb) when phosphorylated at threonine 356. a post-translational modification critical for regulating cell cycle progression. Rb is a tumor suppressor protein that controls the G1-to-S phase transition by binding and inhibiting E2F transcription factors. Phosphorylation of Rb, mediated by cyclin-dependent kinases (CDKs) such as CDK4/6. disrupts this interaction, releasing E2F to activate genes required for DNA replication. Thr356 phosphorylation occurs during the mid-G1 phase and is a key marker of cell cycle commitment.

This antibody is widely used in cancer research to study Rb inactivation mechanisms, as hyperphosphorylation of Rb (due to CDK dysregulation) is a hallmark of many cancers. It helps assess cell proliferation status, drug responses (e.g., CDK4/6 inhibitors like palbociclib), and Rb signaling dynamics in diseases.
