GCET2 (Germinal Center B-cell Expressed Transcript 2), also known as human germinal center-associated lymphoma (HGAL) protein, is a 28 kDa cytoplasmic protein encoded by the *GCET2* gene located on chromosome 3q13. It is selectively expressed in germinal center (GC) B-cells and plays a role in regulating B-cell receptor signaling, cell motility, and immune synapse formation. GCET2 antibodies are widely utilized as immunohistochemical markers to identify GC-derived B-cell lymphomas, particularly in the classification of diffuse large B-cell lymphomas (DLBCLs) into molecular subtypes (e.g., germinal center B-cell-like [GCB] vs. activated B-cell-like [ABC] subtypes).

The antibody detects GCET2 protein expression in formalin-fixed, paraffin-embedded tissues, aiding in differentiating GC-derived lymphomas from non-GC or post-GC malignancies. Studies show that GCET2 expression correlates with better prognosis in DLBCL when combined with other GC markers like CD10 and BCL6. However, its expression is absent in non-neoplastic plasma cells and most non-GC B-cell lymphomas. As a member of the serine protease inhibitor (serpin) superfamily, GCET2 may also modulate immune responses, though its exact molecular mechanisms remain under investigation. Its specificity makes GCET2 a valuable tool in diagnostic hematopathology and research exploring GC B-cell biology and lymphomagenesis.
