Ribophorin I is a conserved transmembrane glycoprotein predominantly located in the rough endoplasmic reticulum (ER). It forms a heterodimeric complex with Ribophorin II, which is essential for the assembly and function of the oligosaccharyltransferase (OST) complex—a critical enzyme machinery mediating N-linked glycosylation. This post-translational modification involves the transfer of preassembled oligosaccharides to specific asparagine residues on nascent polypeptides, playing a vital role in protein folding, quality control, and cellular trafficking. Ribophorin I also interacts with ribosomes and Sec61 translocons, facilitating the coupling of protein translocation and glycosylation.
