Loricrin is a key structural protein essential for the formation of the epidermal cornified envelope, a critical component of the skin barrier. Expressed predominantly in the stratum granulosum of the epidermis, loricrin constitutes ~70% of the total protein mass in the cornified envelope. During terminal differentiation, it undergoes cross-linking with other proteins (e.g., involucrin, small proline-rich proteins) via transglutaminase-mediated bonds, contributing to the mechanical resilience and waterproofing properties of the outermost skin layers. This process is vital for protecting against environmental stressors, pathogens, and water loss.

Loricrin antibodies are immunological tools used to study epidermal differentiation and skin pathology. These antibodies specifically target loricrin epitopes, enabling researchers to visualize its expression and distribution in tissues through techniques like immunohistochemistry (IHC) or immunofluorescence (IF).
