Destrin, also known as actin-depolymerizing factor (ADF), is a key regulator of actin cytoskeleton dynamics. It belongs to the ADF/cofilin protein family, which severs and depolymerizes actin filaments, enabling rapid cytoskeletal remodeling critical for processes like cell migration, cytokinesis, and synaptic plasticity. Destrin's activity is tightly controlled by phosphorylation; LIM kinase-mediated phosphorylation at Ser-3 inhibits its actin-binding ability, while dephosphorylation by phosphatases like slingshot restores its function. Dysregulation of Destrin is implicated in pathological conditions, including cancer metastasis, neurodegenerative disorders, and immune cell dysfunction.
