VPS26 antibodies are essential tools for studying the VPS26 protein family, which comprises two paralogs, VPS26A and VPS26B. These proteins are key components of the retromer complex, a conserved eukaryotic machinery critical for endosomal protein sorting and trafficking. The retromer, composed of a VPS26-VPS29-VPS35 trimer and sorting nexin proteins, mediates retrograde transport of cargo from endosomes to the trans-Golgi network or plasma membrane, ensuring proper cellular homeostasis. VPS26A is the predominant isoform, widely expressed and involved in recycling receptors like cation-independent mannose 6-phosphate receptor (CI-MPR). VPS26B, though structurally similar, exhibits distinct tissue expression and may regulate specialized trafficking pathways or compensate for VPS26A loss.
