LIMPII (Lysosomal Integral Membrane Protein II), also known as SCARB2 (Scavenger Receptor Class B Member 2), is a transmembrane protein primarily localized in lysosomal and endosomal membranes. Discovered in the late 1980s, it plays a critical role in lysosomal biogenesis, membrane trafficking, and cellular lipid metabolism. Structurally, it contains two large luminal domains and a short cytoplasmic tail, facilitating interactions with intracellular transport machinery.

LIMPII gained attention for its dual role in health and disease. It functions as a receptor for enterovirus 71 (EV71) and coxsackievirus A16 (CV-A16), pathogens linked to hand, foot, and mouth disease. This discovery highlighted its significance in viral pathogenesis and immune responses. Additionally, LIMPII is implicated in lysosomal storage disorders, such as Gaucher disease, where impaired lysosomal function leads to pathological accumulations.

Recent studies explore LIMPII's involvement in neurodegenerative diseases, including Parkinson’s, due to its role in α-synuclein clearance. Its regulatory effects on autophagy and lysosomal degradation pathways further underscore its therapeutic potential. Research continues to investigate LIMPII as a biomarker for lysosomal dysfunction and a target for drug development in viral infections and metabolic disorders.
