The plasma membrane calcium ATPase 1 (PMCA1) antibody is a key tool for studying the PMCA1 protein, a member of the P-type ATPase family responsible for calcium ion transport across cell membranes. PMCA1. encoded by the ATP2B1 gene, plays a critical role in maintaining cellular calcium homeostasis by actively extruding calcium from the cytoplasm to the extracellular space. It is ubiquitously expressed, with particularly high levels in excitable tissues like the brain, heart, and skeletal muscle. PMCA1 consists of 10 transmembrane domains and cytoplasmic N- and C-terminal regions, with functional activity modulated by calmodulin binding and alternative splicing.

PMCA1 antibodies are typically produced in rabbits or mice using immunogenic peptides or recombinant protein fragments corresponding to conserved regions of PMCA1. These antibodies enable the detection and localization of PMCA1 in techniques such as Western blotting, immunohistochemistry, and immunofluorescence. Specificity is crucial, as PMCA isoforms (PMCA1-4) share structural homology. High-quality PMCA1 antibodies are validated using knockout controls or siRNA-mediated protein depletion to confirm target specificity.
