Alpha-internexin, a 66 kDa neuronal intermediate filament (IF) protein, plays a critical role in neuronal development and axon outgrowth. It belongs to the neurofilament family, alongside NF-L, NF-M, and NF-H, but is encoded by a separate gene. Highly expressed in the developing nervous system, alpha-internexin is among the earliest neuronal IF markers, appearing in neuroblasts and progenitor cells during embryogenesis. In mature neurons, it co-assembles with other neurofilaments to form the cytoskeletal framework, influencing axonal caliber and stability.

Antibodies targeting alpha-internexin are essential tools in neuroscience research. They enable the detection and localization of the protein in tissues or cell cultures via techniques like immunohistochemistry, Western blotting, or immunofluorescence. These antibodies have been particularly valuable in studying neurodegenerative diseases, such as Alzheimer’s and Parkinson’s, where disrupted IF networks are implicated. Additionally, alpha-internexin overexpression has been observed in neuroendocrine tumors and gliomas, making its antibodies relevant in neuro-oncology for diagnostic or prognostic assessments.
