The GNAQ antibody targets the G protein subunit alpha q (Gαq), encoded by the GNAQ gene. Gαq is a member of the Gα protein family that mediates signaling through G protein-coupled receptors (GPCRs). Upon receptor activation, Gαq stimulates phospholipase C-beta (PLC-β), triggering downstream pathways like the IP3/DAG cascade, which regulates calcium signaling and protein kinase C (PKC) activation. Dysregulation of GNAQ is linked to various pathologies. Notably, somatic mutations in GNAQ, particularly at codon 209 (e.g., p.Gln209Leu), are oncogenic drivers in uveal melanoma, blue nevi, and other melanocytic tumors. These mutations result in constitutive Gαq activation, promoting uncontrolled cell proliferation and survival via MAPK and Hippo/YAP signaling pathways.

GNAQ antibodies are essential tools in research and diagnostics, enabling detection of Gαq expression or mutation status in tissues or cell lines. They are used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to study Gαq's role in normal physiology and disease. In clinical contexts, detecting mutant Gαq helps stratify patients with uveal melanoma, informing prognosis and emerging targeted therapies. Additionally, GNAQ antibodies aid in exploring therapeutic strategies, such as inhibitors targeting downstream effectors (e.g., PKC or MEK).
