Periostin, a multifunctional extracellular matrix protein encoded by the *POSTN* gene, plays critical roles in tissue development, remodeling, and repair. It is highly expressed in collagen-rich connective tissues, particularly during bone formation, cardiac development, and wound healing. Periostin interacts with integrin receptors and other matrix components to regulate cell adhesion, differentiation, and fibrosis. Dysregulation of periostin has been implicated in pathological conditions such as fibrosis, asthma, atopic dermatitis, cancer metastasis, and cardiovascular diseases, making it a target for therapeutic and diagnostic research.

Antibodies targeting periostin are essential tools for studying its expression, localization, and function. These antibodies are typically developed against specific epitopes within periostin’s four conserved fasciclin-like (FAS1) domains or its EMI domain, which mediate protein-protein interactions. Both monoclonal and polyclonal periostin antibodies are widely used in techniques like immunohistochemistry, Western blotting, and ELISA to investigate periostin’s role in disease models or clinical samples. For example, they help identify periostin overexpression in tumor stroma or elevated serum levels in allergic inflammation.

Clinically, anti-periostin antibodies are explored as potential therapeutics. In asthma and atopic dermatitis, neutralizing antibodies aim to block periostin’s interaction with inflammatory pathways. Similarly, in oncology, they may inhibit periostin-mediated cancer cell survival and metastasis.
