Parvalbumin (PV) is a calcium-binding protein belonging to the EF-hand protein family, widely expressed in the nervous system, muscle tissue, and endocrine cells. First identified in fish muscle, it plays a critical role in calcium buffering and signaling, particularly in fast-twitch muscle fibers and GABAergic interneurons in the brain. In the central nervous system, PV is a marker for a subset of inhibitory interneurons that regulate network oscillations and synaptic plasticity. These fast-spiking PV+ neurons are essential for maintaining excitatory-inhibitory balance and are implicated in cognitive functions, including memory and sensory processing.

PV antibodies are vital tools for identifying and studying these specialized neurons. Generated through immunization with purified PV protein or peptide fragments, these antibodies enable visualization of PV-expressing cells via immunohistochemistry, immunofluorescence, and Western blotting. Their specificity is crucial for mapping neuronal circuits and investigating PV's role in diseases such as epilepsy, schizophrenia, and autism, where PV+ neuron dysfunction is observed. Additionally, PV antibodies aid in studying muscle disorders linked to calcium dysregulation. Recent advances in antibody validation techniques, including knockout controls, have enhanced their reliability in both research and diagnostic applications, solidifying their importance in neuroscience and beyond.
