The Phospho-PKCζ (Thr560) antibody detects the activated form of Protein Kinase C zeta (PKCζ), a member of the atypical PKC subfamily. Unlike conventional PKC isoforms, PKCζ does not require calcium or diacylglycerol for activation but relies on phosphorylation for its enzymatic activity. Thr560. located in the activation loop of the kinase domain, is a critical phosphorylation site required for PKCζ activation. This phosphorylation event, mediated by upstream kinases such as PDK1 (3-phosphoinositide-dependent protein kinase 1), stabilizes the active conformation of PKCζ, enabling its participation in downstream signaling pathways.

PKCζ plays a central role in cell survival, polarity, and inflammatory responses, often interacting with components of the PI3K/Akt/mTOR and NF-κB pathways. Dysregulation of PKCζ activity has been implicated in cancer, insulin resistance, and neurodegenerative disorders. The Phospho-PKCζ (Thr560) antibody is widely used in research to study PKCζ activation dynamics under conditions such as growth factor stimulation, oxidative stress, or metabolic challenges.
