**Background of CTCFL Antibody**

CTCFL (CTCF-like), also known as BORIS (Brother of the Regulator of Imprinted Sites), is a paralog of the chromatin organizer protein CTCF. While CTCF is ubiquitously expressed and regulates 3D genome architecture, transcriptional insulation, and imprinting, CTCFL expression is largely restricted to germ cells and certain cancers. CTCFL shares DNA-binding specificity with CTCF but exhibits distinct roles in epigenetic reprogramming, particularly during gametogenesis and oncogenesis.

Antibodies targeting CTCFL are critical tools for studying its unique functions in germ cell development, cancer biology, and epigenetic regulation. CTCFL is aberrantly activated in various cancers (e.g., melanoma, breast, and lung cancers), where it may antagonize CTCF-mediated tumor suppression, alter chromatin states, and promote oncogenic gene expression. Its role in testicular germ cell tumors and germline-specific imprinting further underscores its biological significance.

CTCFL antibodies enable detection of its expression patterns, chromatin localization (via ChIP-seq), and interactions with other proteins. They help distinguish CTCFL from CTCF in co-expression studies, given their overlapping DNA motifs but divergent biological contexts. Research using these antibodies has advanced understanding of CTCFL’s contribution to cancer plasticity, metastasis, and epigenetic instability, positioning it as a potential therapeutic target. However, challenges remain in elucidating its regulatory mechanisms and tissue-specific roles.
