Upon Hh binding, PTCH1-mediated inhibition is relieved, activating downstream transcription factors that regulate embryonic development, tissue homeostasis, and cell proliferation. Dysregulation of PTCH1 is linked to cancers, including basal cell carcinoma and medulloblastoma, often due to inactivating mutations or deletions.
