Desmocollin-3 (DSC3) is a member of the desmocollin family, calcium-dependent transmembrane glycoproteins that are critical components of desmosomes—specialized cell-cell adhesion structures in epithelial and other tissues. As a key desmosomal cadherin, DSC3 mediates intercellular adhesion by binding to desmogleins and linking intermediate filaments to the plasma membrane, thereby maintaining tissue integrity and mechanical resilience. Dysregulation of DSC3 expression has been implicated in several pathological conditions. In cancer biology, DSC3 exhibits dual roles: it may act as a tumor suppressor in certain contexts (e.g., squamous cell carcinomas) by promoting cell adhesion, while its overexpression in other malignancies (e.g., invasive breast cancer) correlates with aggressive phenotypes. Additionally, DSC3 mutations are associated with inherited skin disorders like hypotrichosis and pemphigus.

DSC3-specific antibodies are widely used in research to investigate epithelial barrier function, tumor progression, and desmosome-related diseases. Common applications include immunohistochemistry (IHC), immunofluorescence (IF), and Western blotting (WB) to assess DSC3 localization and expression levels. Commercially available antibodies often target extracellular or intracellular domains of DSC3. with validation in specific experimental models required due to potential cross-reactivity with other desmocollin isoforms. Recent studies also explore DSC3's role in signaling pathways and its potential as a diagnostic or therapeutic target in epithelial cancers.
