CITED2 (Cbp/p300-interacting transactivator with Glu/Asp-rich carboxy-terminal domain 2) is a transcriptional co-regulator belonging to the CITED family. It interacts with the CBP/p300 coactivators, which are critical for mediating transcriptional activation by binding to transcription factors and chromatin remodeling complexes. CITED2 is known to modulate diverse cellular processes, including hypoxia response, embryonic development, cell differentiation, and apoptosis. Its role in hypoxia signaling involves competing with HIF-1α for binding to CBP/p300. thereby fine-tuning the transcriptional response to low oxygen levels.

In development, CITED2 is essential for heart morphogenesis, neural crest cell migration, and left-right patterning. Knockout studies in mice reveal embryonic lethality due to cardiac and adrenal defects. It also influences stem cell pluripotency and tissue-specific differentiation. Dysregulation of CITED2 has been implicated in cancers, cardiovascular diseases, and metabolic disorders, with its function context-dependent—acting as either an oncogene or tumor suppressor.

CITED2 antibodies are widely used in research to detect protein expression, localization, and interactions via techniques like Western blotting, immunohistochemistry, and co-immunoprecipitation. They help elucidate CITED2's regulatory mechanisms in development and disease, making them vital tools for studying transcriptional networks and potential therapeutic targets. Validation of antibody specificity remains crucial due to homology among CITED family members.
