The CIITA (Class II Major Histocompatibility Complex Transactivator) antibody is a tool used to study the CIITA protein, a master regulator of MHC class II gene expression. CIITA is essential for adaptive immunity, as it controls the transcription of MHC-II molecules, which are critical for antigen presentation by immune cells like dendritic cells, B cells, and macrophages. Mutations in the *CIITA* gene are linked to hereditary immune disorders, such as Bare Lymphocyte Syndrome Type II (BLS II), characterized by severe immunodeficiency due to absent MHC-II expression.

CIITA functions as a transcriptional coactivator, binding to DNA-bound regulators like RFX and recruiting histone-modifying enzymes to promote chromatin remodeling. Its activity is tightly regulated by cytokines (e.g., IFN-γ) and cell-specific signaling pathways. Antibodies targeting CIITA enable researchers to investigate its expression, localization, and interactions in immune responses, autoimmune diseases (e.g., rheumatoid arthritis), and cancer. In oncology, CIITA’s role in tumor immunogenicity is of particular interest, as its dysregulation may impair antitumor immunity. These antibodies are widely used in techniques such as Western blotting, immunofluorescence, and ChIP-seq, providing insights into MHC-II-related pathologies and therapeutic strategies.
