IFI16 (Interferon Gamma Inducible Protein 16) is a member of the PYHIN protein family, primarily involved in innate immune responses. It functions as a nuclear DNA sensor, detecting pathogenic or mislocalized DNA from viruses, bacteria, or damaged cells, thereby triggering interferon and pro-inflammatory cytokine production. Structurally, IFI16 contains two HIN-200 domains for DNA binding and a pyrin domain for protein-protein interactions.

Research highlights its dual role in antiviral defense and autoimmunity. It restricts viral replication (e.g., HSV, HPV) by modulating transcription or promoting chromatinization. However, aberrant IFI16 activity is linked to autoimmune diseases like systemic lupus erythematosus (SLE) and Sjögren’s syndrome, where autoantibodies against IFI16 correlate with disease severity.

IFI16 antibodies are essential tools for studying its expression, localization, and interactions in immune signaling pathways.
