The RTN4R antibody targets the Reticulon 4 Receptor (RTN4R), also known as the Nogo Receptor, a glycoprotein predominantly expressed in the central nervous system (CNS). RTN4R plays a critical role in regulating axonal growth inhibition by binding to myelin-associated inhibitors like Nogo-A, MAG, and OMgp. These interactions activate intracellular signaling pathways, such as RhoA/ROCK, which suppress neuronal regeneration and plasticity. RTN4R is structurally characterized by leucine-rich repeats and belongs to the Nogo receptor family, which includes related proteins like LINGO-1 and NgR2. Its function is closely tied to limiting CNS repair post-injury, as it stabilizes neural circuits by restricting excessive axonal sprouting.

Antibodies against RTN4R are widely used in neuroscience research to study mechanisms of neural regeneration, synaptic plasticity, and CNS injury responses. By blocking RTN4R activity, these antibodies have been shown to enhance axonal regeneration in preclinical models of spinal cord injury and stroke. Additionally, RTN4R is implicated in neurological disorders such as Alzheimer's disease and multiple sclerosis, where its aberrant signaling may contribute to neurodegeneration. Research tools targeting RTN4R, including neutralizing antibodies, remain vital for exploring therapeutic strategies to overcome myelin-mediated inhibition and promote functional recovery in CNS pathologies.
