MRGPRX1 (Mas-related G protein-coupled receptor X1) is a member of the MRGPR family, a subset of G protein-coupled receptors (GPCRs) predominantly expressed in sensory neurons, particularly those with pseudounipolar morphology in dorsal root ganglia (DRG) and trigeminal ganglia. It plays a role in detecting endogenous and exogenous stimuli, including pain, itch, and inflammation. MRGPRX1 is activated by neuropeptides, such as cortistatin and proadrenomedullin peptides, as well as by small molecules like chloroquine. Its activation triggers intracellular signaling pathways (e.g., Gq and Gi/o), leading to neuronal excitation and neuropeptide release, which modulate immune responses and sensory transmission.

Antibodies targeting MRGPRX1 are critical tools for studying its expression, localization, and function. They enable visualization of receptor distribution in tissues and help dissect its role in pathological conditions, such as chronic itch, neuropathic pain, and neurogenic inflammation. Recent studies highlight MRGPRX1's involvement in mast cell activation and cross-talk with immune cells, suggesting therapeutic potential in allergic dermatitis or autoimmune disorders. However, challenges remain due to receptor diversity across species (e.g., absent in rodents) and overlapping ligand specificity within the MRGPR family. MRGPRX1-specific antibodies aid in developing targeted therapies while minimizing off-target effects, offering avenues for precision medicine in sensory and immune-related diseases.
