TRPM1 (Transient Receptor Potential Melastatin 1) is a calcium-permeable ion channel belonging to the TRP superfamily, primarily expressed in melanocytes, retinal bipolar cells, and melanoma cells. It plays critical roles in melanin synthesis, retinal signaling, and cellular responses to environmental stimuli. TRPM1 dysfunction is linked to congenital stationary night blindness (CSNB) due to its role in ON-bipolar cell depolarization, as well as melanoma progression, where its expression often inversely correlates with tumor aggressiveness.

TRPM1 antibodies are immunological tools designed to detect and study this protein in research and diagnostic contexts. These antibodies are widely used in techniques like immunohistochemistry (IHC), Western blotting, and immunofluorescence to map TRPM1 expression patterns in tissues, investigate its regulation in melanoma, and explore its pathophysiology in ocular disorders. Commercially available antibodies typically target specific epitopes in the intracellular N-terminal or C-terminal regions of TRPM1. though validation remains essential due to occasional cross-reactivity with other TRP channels.

In clinical diagnostics, TRPM1 antibodies aid in differentiating melanoma subtypes and assessing metastatic potential. Recent studies also explore their potential in developing targeted therapies. However, challenges persist in standardizing detection protocols, as TRPM1 exhibits complex post-translational modifications and tissue-specific splice variants. Ongoing research continues to refine antibody specificity and expand applications in both basic science and translational medicine.
