The CLDN5 antibody is a crucial tool for studying claudin-5 (CLDN5), a member of the claudin family of transmembrane proteins that form tight junctions between epithelial and endothelial cells. Primarily expressed in vascular endothelial cells, CLDN5 is a key component of the blood-brain barrier (BBB), regulating paracellular permeability and maintaining homeostasis in the central nervous system. Its selective expression in the BBB makes it a biomarker for studying neurological disorders, including stroke, Alzheimer’s disease, and brain tumors. Dysregulation of CLDN5 has also been linked to cancer metastasis, as altered tight junction integrity may facilitate tumor cell invasion.

CLDN5 antibodies are widely used in immunohistochemistry (IHC), immunofluorescence (IF), and Western blotting to visualize protein localization, expression levels, and structural integrity in tissue samples or cultured cells. These antibodies aid in assessing BBB disruption in neurodegenerative diseases or evaluating vascular changes in tumors. Researchers also utilize CLDN5 antibodies in functional studies to explore its role in cell-cell adhesion, barrier formation, and signaling pathways. Commercially available antibodies are typically raised against specific epitopes of human or murine CLDN5. with validation across multiple applications. However, cross-reactivity with other claudin family members (e.g., CLDN3 or CLDN12) remains a consideration, necessitating careful validation. Overall, CLDN5 antibodies are indispensable for advancing research in neurobiology, oncology, and vascular biology.
