The PRKCSH antibody targets the protein encoded by the PRKCSH gene, also known as glucosidase II subunit beta (GIIβ). This gene encodes a regulatory subunit of glucosidase II, an enzyme critical for N-linked glycan processing in the endoplasmic reticulum (ER). Glucosidase II trims glucose residues from glycoproteins during protein folding, a step essential for quality control and proper trafficking of secretory proteins. PRKCSH mutations are linked to autosomal dominant polycystic liver disease (ADPLD), a disorder characterized by numerous hepatic cysts. Researchers utilize PRKCSH antibodies to study its expression, localization, and role in disease mechanisms, particularly in cystogenesis.

In ADPLD, PRKCSH loss-of-function mutations disrupt glucosidase II activity, leading to defective glycoprotein maturation, ER stress, and aberrant cell proliferation. PRKCSH antibodies are vital tools in Western blotting, immunohistochemistry, and immunofluorescence to assess protein levels in tissues or cell models. They also aid in exploring interactions with other ER-resident proteins, such as glucosidase II alpha subunit (GIIα), to elucidate functional complexes. Additionally, these antibodies contribute to diagnostic research, helping differentiate PRKCSH-related cystic disorders from other hepatic or renal cystic diseases.
