ERLIN1 (ER lipid raft-associated 1) is a transmembrane protein localized to the endoplasmic reticulum (ER) membrane, belonging to the SPFH (stomatin/prohibitin/flotillin/HflK/C) domain-containing protein family. It plays a critical role in ER-associated degradation (ERAD), a process that targets misfolded proteins for proteasomal degradation. ERLIN1 forms a complex with ERLIN2. which facilitates the recognition and ubiquitination of specific ERAD substrates, such as activated inositol 1.4.5-trisphosphate receptors (IP3Rs). This interaction is crucial for maintaining ER homeostasis and cellular stress responses. Additionally, ERLIN1 is implicated in regulating cholesterol metabolism and apoptosis by modulating sterol-regulated degradation of HMG-CoA reductase and influencing caspase activation.

Antibodies targeting ERLIN1 are essential tools for studying its expression, localization, and molecular interactions. They are widely used in techniques like Western blotting, immunoprecipitation, and immunofluorescence to investigate ERLIN1's role in ERAD, lipid raft organization, and disease contexts, including cancer and neurodegenerative disorders. Dysregulation of ERLIN1 has been linked to breast cancer progression and hereditary spastic paraplegia, underscoring its biomedical relevance.
