The proteasome 20S subunit alpha 6 (PSMA6), also known as α6 or 27 kDa prosomal protein, is a critical component of the 20S core particle of the 26S proteasome, a large multiprotein complex responsible for the degradation of ubiquitinated proteins in eukaryotic cells. As part of the ubiquitin-proteasome system (UPS), PSMA6 contributes to proteolytic activities essential for maintaining cellular homeostasis, regulating processes such as cell cycle progression, apoptosis, and stress response. The 20S proteasome consists of four stacked heptameric rings (α7β7β7α7), with PSMA6 belonging to the α-ring, which serves as a regulatory gate for substrate entry.

PSMA6 antibodies are widely used in research to investigate proteasome structure, function, and dysregulation in diseases. These antibodies enable the detection and quantification of PSMA6 expression in tissues or cells via techniques like Western blotting, immunohistochemistry, and immunofluorescence. Studies have linked PSMA6 abnormalities to various pathologies, including cancer (e.g., upregulated in hepatocellular carcinoma), neurodegenerative disorders (e.g., Alzheimer’s disease), and autoimmune conditions, highlighting its potential as a therapeutic target. Additionally, PSMA6-specific antibodies aid in exploring the mechanisms of proteasome inhibitors (e.g., bortezomib) used in cancer therapy.
