The HBZ (HTLV-1 bZIP factor) antibody is associated with research on human T-cell leukemia virus type 1 (HTLV-1), a retrovirus linked to adult T-cell leukemia/lymphoma (ATLL) and HTLV-1-associated myelopathy. The HBZ gene, encoded by the antisense strand of the HTLV-1 provirus, produces two isoforms: a spliced mRNA (HBZ-SI) and an unspliced form (HBZ-US). The HBZ protein interacts with host cellular pathways, notably modulating transcription factors like CREB and NF-κB, which promote viral persistence and T-cell proliferation while suppressing viral replication. This dual role makes HBZ critical for HTLV-1 pathogenesis and immune evasion.

HBZ antibodies are primarily studied to understand immune responses in HTLV-1-infected individuals. While most HTLV-1 research focuses on structural proteins (e.g., Tax), HBZ-specific antibodies have gained attention due to their potential as biomarkers. Studies suggest HBZ antibody detection in patient sera correlates with disease progression, particularly in ATLL. However, HBZ's weak immunogenicity and low expression levels pose challenges for antibody detection, requiring highly sensitive assays like luciferase immunoprecipitation systems (LIPS).

Recent efforts aim to optimize HBZ antibody assays using recombinant proteins or peptide-based platforms. These advancements could improve diagnostic accuracy and provide insights into HTLV-1 latency mechanisms. Despite progress, the clinical utility of HBZ antibodies remains under investigation, with ongoing studies exploring their prognostic value and role in immune surveillance.
